When I turn computer on I hear giant pitched clap, won,t boot up for 5 min, afterwards starts , whats the problem?

This have been up for a while but is slowly getting worse. Monitor goes into standby mode in the past it boots up as well. I ,ve be told by more than one individual its either the thorny drive at fault or something contained by the motherboard. Is it a minor problem or very serious??
